Our project focuses on bone cell interactions that are implicated in bone pathologies. Based on our previous work, we study bone cells and their communications through soluble mediators that appeared to play an important role in bone healing, inflammatory arthritis and primary bone tumors. Our project aims:

  • To characterize the interactions between stromal cells (mesenchymal stem cells, fibroblasts, osteoblasts) and myeloid cells (monocytes, macrophages, osteoclasts) in bone healing or inflammatory arthritis.
  • To model cell interactions in primary bone tumors by using co-cultures and 3D bioprinting.
  • To propose new therapies for bone pathologies including primary bone tumors based on these cell interactions.
Our team gathers 3 researchers with complementary expertise (F. Blanchard, P. Layrolle and V. Trichet) and clinicians from the academic hospital of Nantes and Tours (B. Le Goff, rheumatologist; P. Perrot, plastic surgeon; P. Rosset, orthopedic surgeon).

Axis 1. Stromal and myeloid cells in bone healing.

In vitro and in vivo study as well as clinical trials on the stromal cells (mesenchymal stem cells, MSC) able to induce bone regeneration: heterogeneity (sampling site, different patients); paracrine effects, secretome and exosomes; communications with myeloid cells (macrophages and osteoclasts); reconstruction of large bone defects with an anatomic pre-vascularized bone transplant; 3D printing of bone substitutes.

In vitro and in vivo study of communications between myeloid (macrophages, osteoclasts) and stromal (synovial fibroblasts, osteoblasts) cells: role and mechanisms of action of novel cytokines from IL-1 family that are produced by macrophages in rheumatoid arthritis; impact and secretome of tumor infiltrating macrophages (TAM) in the development of bone sarcomas (osteosarcoma, Ewing sarcoma).

In vitro and in vivo study of stromal cells in bone sarcomas: mesenchymal stem cells (MSC) versus cancer stem cells (CSC); genomic, phenotypic and metabolic characteristics; supporting role of MSC on tumor development; tumorigenicity in SCID mice; 3D modeling in co-cultures of cancer cells with bone cells on biomimetic matrix; 3D bioprinting of tumors; organotypic slices of bone tumors.
